Comparison of Autofluorescence and pH Changes in Acinetobacter baumannii and Candida albicans Cultures based on Incubation Time

Abstract

|
Objectives: Quantitative Light-induced Fluorescence-Digital (QLF-D) was used to investigate the relationship between the autofluorescence characteristics of Acinetobacter baumannii and Candida albicans (a major causative agent and a risk factor for nosocomial pneumonia, respectively) and pH change.
Methods: A. baumannii and C. albicans were the experimental microorganisms, and Streptococcus mutans was the control. Autofluorescence expression was assessed by smearing the cells on a solid medium and imaging for QLF-D at 48 h intervals for 168 h. The R/G ratio was calculated for the three types of colonies. The pH of the cultures (in liquid media) was measured for each incubation time. For statistical analysis, the Kruskal-Wallis test and Pearson correlation analysis were performed using SPSS version 25.0.
Results: A. baumannii emitted orange fluorescence; the R/G ratio and pH increased at 24 h and peaked at 168 h, and exhibited positive correlation. C. albicans emitted reddish fluorescence; the R/G ratio and pH increased at 24 h, peaked at 120 h, and subsequently decreased. S. mutans did not exhibit fluorescence.
Conclusions: The fluorescence of the A. baumannii cells was clearly observed, and the pH of the medium increased. The pH of the C. albicans culture peaked at 120 h and then decreased gradually. The correlation between autofluorescence and pH was confirmed.
